Exploring the principles of microscopy, this guide covers magnification calculations, relationships between magnification and field of view at different powers, sizing objects within the field of view, and determining total magnification in drawings. Visual aids enhance the understanding of these concepts and calculations.

  1. Microscope Lab Magnification in the microscope Ocular lens(X) times Objective lens(X) = total magnification(X) 1. 10X times 40X = 400X 2. 10X times 100X = 1000X

  2. Microscope Lab Field of view The total size of the image that you re viewing e.g. the field of view at low power is about 3.6 mm (note lab book says cm and is wrong) 1 micrometer ( m) = 1/1,000,000 m or 10-6m 1 millimeter (mm) = 1/1000 m or 10-3m 3.6 mm = 3600 m

  3. Microscope Lab Field of view is directly proportional to magnification 3. The field of view at low power (50X) is about 3.6 mm. What is the field of view at medium power (100X)? 3.6/2 = 1.8 mm or 1800 m 4. The field of view at medium power (100X) is about 1.75 mm. What is the field of view at high power (400X)? 1.75/4 = 0.4375 mm or 437.5 m What is the field of view at oil immersion power (1000X)? 1.75/10 = 0.175 mm or 175 m

  4. Microscope Lab Size of objects in field of view 5. The field of view at medium power (100X) is about 1.75 mm. Seven objects can fit across the field of view. How big is the object? 1.75/7 = 0.25 mm

  5. Microscope Lab Size of objects in field of view 6. The field of view at medium power (100X) is about 1.75 mm. At high power (400X), you estimate that 12 objects fit across the field of view. Find field of view at high power first 1.75/4 = 0.4375 mm or 437.5 m Then find the size of the object 437.5 m/12 = 36.46 m

  6. Microscope Lab Total magnification of a drawing 8. You estimate the size of the object is 150 m. You produce a drawing 10 cm across. What magnification is the drawing First, use common units: 10 cm = 100,000 m Then find the magnification: 100,000 m/150 m = 667

  7. Microscope Lab 8. Total magnification of a drawing The field of view is 3.6 mm at medium power (50X). Five objects can fit across the field of view at high power (400X). Your drawing shows one object, 27 cm across. field of view at high power = 3.6/8 = 0.45 mm 0.45/5 = 0.09 mm 270 mm/0.09 mm = 3000 X
